Exercise #10 – Hypothesis & Experimental Design
Objective: To determine the effect of a chosen solvent on membrane solubility, specifically the tonoplast of beet cells, by quantifying the release of betacyanin.
Possible list of solvents:
Liquids
Methanol
Acetone
Ethanol
Bleach
Ethyl acetate
Salts – Must be below 25% concentration
NaCl
CaCl2
NaF
NaHCO3
CuSO4 · 5H2O
MgO
Acids – Must be below 30%
HCl
H2SO4
HNO3
Hypothesis & Experimental Design
– Select one of the chemicals above to be used as a solvent to test the quantity of betacyanin released using a spectrophotometer. The concentrations you choose must be based on scientific literature or your textbook/manual.
Structure of Assignment:
– Provide a one paragraph justification for using the solvent (based on current literature and textbooks). You must cite within your paragraph. This will serve as part of your introduction
– Objective (one sentence)
– Hypothesis: Null & Alternative (one sentence each)
– Experimental Plan – Here, list the following components
o Four concentrations of the solvent to be tested, including a control. Concentrations can be in % or molarity.
o List of all concentration treatments with the mass needed per liter of solution (or volume needed per liter) i.e., 2mM treatment = 2 g/l
o Length of experiment
o Number of replicates: three
o Volume of each: 5 ml
o Data collected: absorbance of solutions at 460 nm
– Bibliography
o Citations using the format included below.
These italicized components should be written as is.
Citations
Citations document the source of statistics, quotations, facts, and ideas that you use in your paper but have gotten from other sources. In ANY academic writing, you must acknowledge and cite your sources. Just to make things more difficult, there are different citation styles for different academic disciplines. Healey Library has an excellent manual that is available to any student. Just pick up a copy at the librarian’s desk in the main fourth floor room of the library. This manual will show you how to format a proper bibliographic citation.
The full description of your source should come at the end of the paper in a list of references or a bibliography. A full description of a source will include:
 the author’s name(s),
 the title of the work (or web page) cited,
 the title of the journal, book, or name of the website (not the web address!) that the work is part of (if the work is an article, chapter of a book, or page on a website),
(NOTE: The three items above should ALWAYS come first)
 the volume/issue number of the journal (if appropriate)
 the location and name of the publisher (for books)
 the year the work was published (in the case of web pages, look for day/month/year “creation date”)
 the URL (the address of the specific web page you are citing)
 the page number (for specific quotes)
 the date you accessed the web page
For some sources you won’t be able to find all this information, but you should be suspicious of sources that are hard to cite!
